Pharmaceutical Suspensions: From Formulation Development to Manufacturing

Pharmaceutical Suspensions: From Formulation Development to Manufacturing.
By Alok K. Kulshreshtha, Onkar N. Singh, G. Michael Wall

  • Publisher: Springer
  • Number Of Pages: 323
  • Publication Date: 2009-10-16
  • ISBN-10 / ASIN: 1441910867
  • ISBN-13 / EAN: 9781441910868
Product Description:

The suspension dosage form has long been used for poorly soluble active ingredients for various therapeutic indications. Development of stable dispersions over the shelf life of the drug product continues to be a challenge on many fronts.

A good understanding of fundamentals of disperse systems is essential in the development of a suitable pharmaceutical suspension. The development of suspension dosage form follows a very complicated path. The selection of the proper excipients (surfactants, viscosity imparting agents etc.) is important. The particle size distribution in the finished drug product dosage form is a critical parameter that significantly impacts the bioavailability and pharmacokinetics of the product. Appropriate analytical methodologies and instruments (chromatographs, viscometers, particle size analyzers, etc.) must be utilized to properly characterize the suspension formulation. The development process continues with a successful scale-up of the manufacturing process. Regulatory agencies around the world require clinical trials to establish the safety and efficacy of the drug product. All of this development work should culminate into a regulatory filing in accordance with regulatory guidelines. Pharmaceutical Suspensions, in its organization, follows the development approach used widely in the pharmaceutical industry. The primary focus of this book is on the classical disperse system – poorly soluble active pharmaceutical active dispersed in a suitable vehicle.

After discussing various disperse systems, the theory of disperse systems and commonly used excipients, the remaining chapters in this textbook systematically detail the development of pharmaceutical suspensions, from pre-formulation stage to clinical development, regulatory submission and commercial manufacturing. Additionally, the emerging area of nano-suspensions as applied to pharmaceutical filed is also discussed. Each of the chapters in Pharmaceutical Suspensions was written independently by scientists skilled in their specific areas. Contributing authors represent a cross-sections of scholars from Academic Institutions, Pharmaceutical Industries and Regulatory Agency.

From the Back Cover In order to reach its intended site of action, the drug molecules in every pill that we swallow must first be absorbed, transported via the bloodstream and evade various mechanisms that eliminate drugs from the body. Those drug properties that determine, for example, its stability in the gut or its ease of uptake into the bloodstream, are therefore of central importance in drug development. In fact, many potentially useful drugs fail because of insufficient availability at the biological target site.

BooK Description
This work presents the chemistry of functional groups with an emphasis on patterns of reactivity, the consequences of the relative electronegativity of the atoms that constitute the functional groups, the role or lone pairs and the stereochemistry of reactions at a particular group. The material is presented in four sections. The first section considers functional groups involving C-X bonds such as alkyl halides, alcohols, ethers and amines. The second section considers unsymmetrical p=bonded C=X functional groups, typically the carbonyl group and the influence of these groups on adjacent centres. The third section covers symmetrical p-bonded functional groups including alkenes, alkynes and arenes and the consequences of the factors that perturb the symmetry, such as on the orientation of aromatic substitution. The final section concentrates on interconversions like oxidation and reduction, and the protection of functional groups to achieve selectivity. The chemistry of the functional groups is considered here with mechanistic rationale. Students will be encouraged to consider reactions in terms of regions of electron deficiency and excess, and hence identify the sites at which electrophiles and nucleophiles might react.

Organic Chemistry

Organic Chemistry.
Publisher: Thomson Brooks/Cole | ISBN: 0534373666 | edition 1999 | DJVU | 1338 pages | 13,57 mb

Widely and consistently praised as the most clearly written book on the market. Why? In John McMurry's words: "I have been asked hundreds of times over the past ten years why I wrote this book. I wrote this book because I love writing. I get great pleasure and satisfaction from taking a complicated subject, turning it around until I see it clearly from a new angle, and then explaining it in simple words. I write to explain chemistry to students the way I wish it had been explained to me years ago." Through his lucid writing and ability to show the beauty and logic of organic chemistry, McMurry makes learning enjoyable for students. The highest compliment that can be given to a chemistry book applies to McMurry: It works! Mainstream in level, McMurry's coverage is concise yet doesn't omit any key topics. McMurry blends the traditional functional-group approach with a mechanistic approach. The primary approach, by functional group, begins with the simple and progresses to the more complex so that readers who are not yet versed in the subtleties of mechanisms are first exposed to the what of chemistry before beginning to grapple with the why. Within this primary organization, the author places a heavy emphasis on explaining the fundamental mechanistic similarities. Currently in use at hundreds of colleges and universities throughout the United States and Canada, McMurry's Organic Chemistry is also an international bestseller from the United Kingdom to the Pacific Rim.

The End of Stress As We Know It By Bruce McEwen, Elizabeth Norton Lasley, Elizabeth Lasley

The End of Stress As We Know It By Bruce McEwen, Elizabeth Norton Lasley, Elizabeth Lasley.
Publisher: National Academies Press 2002-10-15 | 285 Pages | ISBN: 0309077201 | PDF | 4.56 MB

There’s a whole new way to think about stress. Sure, some stress is inevitable, but being “stressed out” isn’t. In fact, we can learn to rechannel the powerful stress activators in our lives to make us even more effective.
Hamlet spoke of “suffering the slings and arrows of outrageous fortune.” These days we simply use the word “stress” to describe that feeling. And if you ask 10 random people if they feel stressed, chances are that at least 9 will reply with a resounding, “Yes!” Indeed, the very way we use the word implies that we are its victims—as in, “I’m under so much stress” or “I’m completely stressed out.” There’s now a better way to look at this picture, a way to move from victim to victor. The first step is to look to the science behind it all because in the science lies a whole new message about stress. Science allows us to understand what the stress response is and why our bodies react the way they do. Like all living creatures, we’re mapped to respond instinctually in certain ways, and generally for good reasons. We know, for example, that in times of emergency, we effortlessly shift into a different biological mode. Based on our perception of the crisis, our brains initiate the “stress response” or the “flight-or-fight reaction.” Our attention becomes keenly focused. Our heart and lungs accelerate to ready us for action. Our glands mobilize extra energy resources and summon the immune system to battle stations. This whole process is Nature’s way of empowering us to respond swiftly, sometimes dramatically, to sudden events, while remaining mentally alert and physically prepared to meet a challenge.

The term "incompatibilities" describes a wide range of chemical reactions that produce undesirable results in noncontrolled situations: the generation of toxic gases, fire, explosions, corrosive activity, polymerization, ruptured containers, creation of more dangerous compounds, and the like. A portable and easy-to-use reference on reactive substances commonly found in commerce, the Wiley Guide to Chemical Incompatibilities, Third Edition compiles hard-to-find data on over 11,000 chemical compounds, providing chemists, technicians, and engineers a thorough, lightning-quick resource to use during experimental preparation and in the event of an emergency.

More than a revision of the previous edition, this Third Edition has been rewritten and expanded to broaden coverage and improve its usefulness. It contains nearly 9,000 chemical incompatibility profiles and nearly 250 new entries, covering flammability, violent and explosive binary reactions, incompatibilities, and reactions that may result from physical change. Alphabetical organization provides concise incompatibility profiles for thousands of commonly used commercial chemicals, allowing readers to look up a given substance and instantly learn whether it is incompatible with common materials, other chemical substances, structural materials, or personal protective equipment.

New for the Third Edition:

  • Chemicals that have the potential to cause disasters
  • Chemical formulas and autoignition temperatures
  • More flash points, as well as molecular formulas, lower and upper explosive limits, autoignition temperatures, and NFPA®-type (Red) numerical fire codes
  • Safety reminders
  • All entries keyed by CAS numbers to eliminate possible confusion among synonyms
  • Spanish-, French-, and German-language entries for international use
  • Revised glossary helps users who may not be chemists with general chemical terms
With thousands of new entries and easy-to-use organization, the Third Edition of the Wiley Guide to Chemical Incompatibilities remains a handy resource for all safety, first-response, and plant management professionals responsible for the handling, storage, and conveyance of chemical materia.
