Preface and Introduction
The publication of this book appears at a time when many colleagues who are
actively involved in analytical research, consider thin-layer chromatography as one
of the less important analytical tools. Another considerable group of scientists
have concentrated their efforts on the problems of HPLC, a method which is called
high pressure liquid chromatography but should be high performance liquid chromatography.
The separation columns - rarely operated under optimum conditions -
hardly exceed lo00 theoretical plates. Irreversible adsorption of sample components
on the column material, often limits their general application. Most of the shortcomings
of HPLC can be avoided by using TLC methods.
This book is not intended to discredit HPLC. Indeed, it is meant for those who
use HPLC. An elegant, fast, inexpensive and sufficiently precise preliminary technique
for HPLC can now be provided which may be the method of choice for
certain analytical problems. The technique has been named HPTLC, high performance
thin-layer chromatography and the editors are fully aware of the implication
of this notation. The following list of characteristics demonstrates the
potential of HPTLC.
Separation capability: up to 40 different substances, completely separated in one
single run.
separation of more than 5 components per minute (time
averaged).
- HPTLC as a preliminary technique for HPLC: analysis
of one sample or a group of substances.
- HPTLC as a routine technique: analysisof up to 12 samples
in one single run using the micro-circular technique;
analysis of up to 8 samples with the macro-circular technique
and up to 40 samples in the linear technique. Using the
continuous flow method, more than 10 samples can be
analyzed successively.
Near a Rf-value range of 0.5, real Rf-values with relative
standard deviations of f 1% can be obtained when the
circular technique (Uchamber-CAMAG) is applied.

Quantitative analysis: Relative standard deviations of+ 2.3% or better are obtained
with the linear technique in the nanogram range. The

During the last decade there has been a steady increase in the use of radiotracers in
organic and biological chemistry and this has been accompanied by continued advances in
the efficiency and sophistication of chromatographic methods. As a result, the chromatographic
and electrophoretic separation of radiolabelled compounds, referred to as “radiochromatography”,
is now carried out in many laboratories throughout the world.
The aim of this book is to describe and discuss the various radiochromatography and
radioelectrophoresis methods in a single volume. The historical development of the techniques
is outlined and the reasons for choice of a method for any particular application
are discussed. This is followed by a more detailed consideration of the practical aspects
and examples of applications taken from the recent literature.

book, particularly Alan Dutton for help with parts of Chapter 6 on radio-high-performance

Analytical Profiles of Drug Substances

The concept for gathering together and publishing pertinent information on the physical and chemical properties of various official and new drug substances had its origin with the members of the Section on Pharmaceutical Analysis and Quality Control of the Academy of Pharmaceutical Sciences.More than two years of consideration preceded the authorization of this ambitious project by the Executive Committee of the Academy in the Spring of 1970. The immediate and virtually spontaneous enlistment of the first group of contributors to this work attested to its importance and the wisdom of pursuing its publication. By coincidence, the delegates to the sesquicentennial anniversary meeting of the United States Pharmacopeial Convention, Inc., in Washington, D. C . on April 8-1 0, 1970, adopted the following resolution: Whereas widespread interest has been expressed in the inclusion of additional information about physical and chemical properties of drugs recognized in the United States Pharmacopeia Be I t Resolved that the Board of Trustees consider publishing in the Pharmacopeia, or in a companion publication, information on such attributes as solubilities, pH and pK values, spectra and spectrophotometric constants, and stability data, pertaining to pharmacopeial drugs.The U.S.P.C. Board of Trustees unanimously approved the resolution in principle on June 4, 1970 and authorized the Director of Revision t o include in the U S.P. monographs such physical-chemical information as he deemed
proper and also to cooperate with the Academy of Pharmaceutical Sciences to secure the publication of other physical-chemical data.It was my privilege to be the President of the Academy during the period when Analytical Profiles was under consideration. It is my unusual
and unique honor as President of the Academy and Director of U.S.P Revision to assist in the institution and dedication of this first volume. I trust that it will serve immeasurably in providing the scientific community with an authoritative source of information on the properties of many o f our important drug compounds.
